Adrenomedullin
Adrenomedullin (ADM) is an endogenous vasodilatory peptide that consists of 52 amino acids and has a wide range of diverse effects and functions. ADM and its receptors are widely distributed in various tissues and are highly expressed in blood vessels. It belongs to the calcitonin gene-related peptide (CGRP) superfamily of vasoactive peptide hormones, and plays a pivotal role in various regulatory mechanisms, particularly those of the cardiovascular and lymphatic system.
ADM exerts biological effects by activation of the calcitonin receptor-like receptor (CLR) as a complex with one of three receptor activity-modifying proteins (RAMP), which alter the ligand affinity. It is seen as a positive influence in diseases such as hypertension, myocardial infarction, chronic obstructive pulmonary disease and other cardiovascular diseases, whereas it can be seen as a negative factor in potentiating the ability of cancerous cells to extend their blood supply and thereby enable further cell proliferation.
